In the synthesis of active pharmaceutical ingredients (APIs), acetates operate at the intersection of mild bases, phase-transfer agents, and coordination chemistry modifiers.

Acid scavenger in API synthesis

During condensation, coupling, or halogenation reactions in API synthesis, hydrochloric or hydrobromic acid is frequently generated as a by-product.

Anhydrous sodium acetate acts as an effective, non-nucleophilic acid scavenger.

It neutralizes the mineral acid to form acetic acid, preventing the degradation of acid-sensitive intermediates without triggering the unwanted side reactions that stronger inorganic bases, such as sodium hydroxide or carbonates, might cause.

Catalysis and cross-coupling reactions

Acetate salts are key components in transition-metal-catalyzed cross-coupling reactions (e.g. Suzuki, Heck, or Buchwald–Hartwig amination). Sodium acetate and potassium acetate frequently serve as bases that facilitate the transmetalation step.
